you could go the way of some elaborate cell shading, the method of making 3d look like hand drawn 2d, but for the most part you'd be losing detail. Its basically just some ramp shaders from what i understand. Really like you're direction (and hair, i've always wanted to have myself anime hair!)

i have boned and skinned and posed her, and let me tell ya, painting the bone weights so she doesnt get all tweaked out of shape when you move the bones around can be tough! took me awhile to figure out how it works anyways, here's some pics of her current state...

oh, a shirt... yea, right..... i guess she would need somethin up there considering how cold it can get on the sea an all. I dunno, we'll see what i can come up with
ok, added some stuff to her: eyeballs, more clothes, arm bands, and boot tops. I think I might try texturing the clothes but Im running out of time and I have a busy weekend ahead of me as it is... anyways, heres a pic of her atm...
